Deciphering the mechanisms that drive the emergence of new phenotypes is a primary aim in biology. Evolutionary changes in regulatory mechanisms, which modify gene expression patterns but leave the gene products intact, are likely responsible for a large fraction of phenotypic innovations. Among the regulatory mechanisms important for phenotypic evolution, those involving non-coding RNAs are of particular interest. It was recently discovered that, in addition to protein-coding genes, mammalian genomes contain thousands of non-coding RNA genes with potential regulatory roles. Notably, long non-coding RNAs (lncRNAs, broadly defined as large RNA molecules lacking protein-coding potential) include regulatory RNAs that played key roles in mammalian evolution. Moreover, lncRNAs evolve rapidly and are largely lineage- specific, thus underlining the need to assess their roles in the emergence of lineage-specific phenotypes.
